This variant lies in the ETFB gene (transcript NM_001985.3) at the canonical splice donor site of the intron immediately after coding-DNA position 375, where G is replaced by A; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: Variant summary: ETFB c.375+1G>A is located in a canonical splice-site and is predicted to affect mRNA splicing resulting in a significantly altered protein due to either exon skipping, shortening, or inclusion of intronic material. Variants that disrupt the consensus splice site are a relatively common cause of aberrant splicing and loss of ETFB function. Several computational tools predict a significant impact on normal splicing: Four predict the variant abolishes a 5' splicing donor site. However, these predictions have yet to be confirmed by functional studies. The variant was absent in 250746 control chromosomes. To our knowledge, no occurrence of c.375+1G>A in individuals affected with Glutaric Aciduria, Type 2b and no experimental evidence demonstrating its impact on protein function have been reported.
