Uncertain significance — the classification assigned by Ambry Genetics to NM_006506.5(RASA2):c.832G>A (p.Val278Ile), citing Ambry Variant Classification Scheme 2023: The p.V278I variant (also known as c.832G>A), located in coding exon 9 of the RASA2 gene, results from a G to A substitution at nucleotide position 832. The valine at codon 278 is replaced by isoleucine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
