Uncertain significance for 3-methylcrotonyl-CoA carboxylase 1 deficiency — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_020166.5(MCCC1):c.1208T>G (p.Val403Gly), citing Invitae Variant Classification Sherloc (09022015): This variant has not been reported in the literature in individuals affected with MCCC1-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This variant is not present in population databases (gnomAD no frequency). This sequence change replaces valine, which is neutral and non-polar, with glycine, which is neutral and non-polar, at codon 403 of the MCCC1 protein (p.Val403Gly).
