Uncertain significance for Wilms tumor 1 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_004484.4(GPC3):c.485A>T (p.Asn162Ile), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces asparagine, which is neutral and polar, with isoleucine, which is neutral and non-polar, at codon 162 of the GPC3 protein (p.Asn162Ile). This variant is present in population databases (no rsID available, gnomAD 0.03%), including at least one homozygous and/or hemizygous individual. This variant has not been reported in the literature in individuals affected with GPC3-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
