Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_005732.4(RAD50):c.2923-11_2923-7del, citing Invitae Variant Classification Sherloc (09022015). This variant lies in the RAD50 gene (transcript NM_005732.4) at 11 bases into the intron immediately before coding-DNA position 2923 through 7 bases into the intron immediately before coding-DNA position 2923, deleting this region. Submitter rationale: This sequence change falls in intron 18 of the RAD50 gene. It does not directly change the encoded amino acid sequence of the RAD50 protein. This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with RAD50-related conditions. ClinVar contains an entry for this variant (Variation ID: 2072564). Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
