Uncertain significance for Charcot-Marie-Tooth disease type 4 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_014845.6(FIG4):c.206G>A (p.Arg69His), citing Invitae Variant Classification Sherloc (09022015): This variant has not been reported in the literature in individuals affected with FIG4-related conditions. This sequence change replaces arginine, which is basic and polar, with histidine, which is basic and polar, at codon 69 of the FIG4 protein (p.Arg69His). This variant is present in population databases (rs748211930, gnomAD 0.009%). Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Tolerated"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C0"). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
