Pathogenic for Beta-D-mannosidosis — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_005908.4(MANBA):c.1534G>T (p.Glu512Ter), citing Invitae Variant Classification Sherloc (09022015): This variant has not been reported in the literature in individuals affected with MANBA-related conditions. For these reasons, this variant has been classified as Pathogenic. This variant is not present in population databases (gnomAD no frequency). This sequence change creates a premature translational stop signal (p.Glu512*) in the MANBA gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in MANBA are known to be pathogenic (PMID: 9384606, 12468273).
