Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_002474.3(MYH11):c.5031G>C (p.Glu1677Asp), citing Ambry Variant Classification Scheme 2023: The p.E1677D variant (also known as c.5031G>C), located in coding exon 34 of the MYH11 gene, results from a G to C substitution at nucleotide position 5031. The glutamic acid at codon 1677 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
