Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_170682.4(P2RX2):c.374G>A (p.Cys125Tyr), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces cysteine, which is neutral and slightly polar, with tyrosine, which is neutral and polar, at codon 125 of the P2RX2 protein (p.Cys125Tyr). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive. This missense change has been observed in at least one individual who was not affected with P2RX2-related conditions (Invitae). This variant has not been reported in the literature in individuals affected with P2RX2-related conditions. The frequency data for this variant in the population databases is considered unreliable, as metrics indicate poor data quality at this position in the gnomAD database.
