Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_005138.3(SCO2):c.479T>G (p.Val160Gly), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces valine, which is neutral and non-polar, with glycine, which is neutral and non-polar, at codon 160 of the SCO2 protein (p.Val160Gly). This variant is present in population databases (rs755433769, gnomAD 0.005%). This missense change has been observed in individual(s) with cardioencephalomyopathy (PMID: 17187620). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. This variant is also known as c.1602T>G. ClinVar contains an entry for this variant (Variation ID: 2066356). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt SCO2 protein function.
