This variant lies in the POLG gene (transcript NM_002693.3) at coding-DNA position 863, where G is replaced by A; at the protein level this means replaces arginine at residue 288 with histidine — a missense variant. Submitter rationale: Variant summary: POLG c.863G>A (p.Arg288His) results in a non-conservative amino acid change located in the DNA mitochondrial polymerase, exonuclease domain (IPR041336) of the encoded protein sequence. Three of five in-silico tools predict a benign effect of the variant on protein function. The variant allele was found at a frequency of 8.2e-06 in 244254 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance.
