Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant is not likely to affect RNA splicing. For these reasons, this variant has been classified as Pathogenic. This premature translational stop signal has been observed in individual(s) with mitochondrial neurogastrointestinal encephalomyopathy (PMID: 10852545, 19748572). This sequence change creates a premature translational stop signal (p.Phe18Leufs*105) in the TYMP gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in TYMP are known to be pathogenic (PMID: 9924029, 15781193). This variant is not present in population databases (gnomAD no frequency).
