Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_018129.4(PNPO):c.698G>A (p.Arg233Gln), citing Ambry Variant Classification Scheme 2023: The p.R233Q variant (also known as c.698G>A), located in coding exon 7 of the PNPO gene, results from a G to A substitution at nucleotide position 698. The arginine at codon 233 is replaced by glutamine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
