Uncertain significance for Maple syrup urine disease — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000709.4(BCKDHA):c.173C>T (p.Ser58Leu), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces serine with leucine at codon 58 of the BCKDHA protein (p.Ser58Leu). The serine residue is highly conserved and there is a large physicochemical difference between serine and leucine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with BCKDHA-related disease. Algorithms developed to predict the effect of missense changes on protein structure and function do not agree on the potential impact of this missense change (SIFT: "Tolerated"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0").
