Uncertain significance for Combined oxidative phosphorylation defect type 17 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_018127.7(ELAC2):c.118C>G (p.Leu40Val), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces leucine, which is neutral and non-polar, with valine, which is neutral and non-polar, at codon 40 of the ELAC2 protein (p.Leu40Val). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with ELAC2-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
