NM_001042492.3(NF1):c.5C>A (p.Ala2Asp) was classified as Uncertain significance for Cardiovascular phenotype; Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.A2D variant (also known as c.5C>A), located in coding exon 1 of the NF1 gene, results from a C to A substitution at nucleotide position 5. The alanine at codon 2 is replaced by aspartic acid, an amino acid with dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
