Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_001277115.2(DNAH11):c.5815G>A (p.Gly1939Arg), citing LabCorp Variant Classification Summary - May 2015: Variant summary: DNAH11 c.5815G>A (p.Gly1939Arg) results in a non-conservative amino acid change in the encoded protein sequence. Three of four in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 4e-06 in 250296 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.5815G>A has been reported in the literature in the compound heterozygous state in trans with a pathogenic variant in an individual affected with Primary Ciliary Dyskinesia 7 (Knowles_2012). These data do not allow any strong conclusion about variant significance. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publications have been ascertained in the context of this evaluation (PMID: 22184204, 34405951, 34133440, 32633470). ClinVar contains an entry for this variant (Variation ID: 2062318). Based on the evidence outlined above, the variant was classified as uncertain significance.
