Uncertain significance for Charcot-Marie-Tooth disease type 2 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001365951.3(KIF1B):c.3785A>G (p.Glu1262Gly), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glutamic acid, which is acidic and polar, with glycine, which is neutral and non-polar, at codon 1216 of the KIF1B protein (p.Glu1216Gly). This variant is present in population databases (rs780214312, gnomAD 0.04%), and has an allele count higher than expected for a pathogenic variant. This variant has not been reported in the literature in individuals affected with KIF1B-related conditions. ClinVar contains an entry for this variant (Variation ID: 2060673). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be tolerated.
