Uncertain significance for Hypertrophic cardiomyopathy; Primary dilated cardiomyopathy — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_014476.6(PDLIM3):c.850G>A (p.Gly284Ser), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the PDLIM3 gene (transcript NM_014476.6) at coding-DNA position 850, where G is replaced by A; at the protein level this means replaces glycine at residue 284 with serine — a missense variant. Submitter rationale: This sequence change replaces glycine, which is neutral and non-polar, with serine, which is neutral and polar, at codon 284 of the PDLIM3 protein (p.Gly284Ser). This variant is present in population databases (rs574011141, gnomAD 0.006%). This variant has not been reported in the literature in individuals affected with PDLIM3-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function output the following: SIFT: "Deleterious"; PolyPhen-2: "Benign"; Align-GVGD: "Class C55". The serine amino acid residue is found in multiple mammalian species, which suggests that this missense change does not adversely affect protein function. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
