This variant lies in the COL7A1 gene (transcript NM_000094.4) at coding-DNA position 1612, where C is replaced by T; at the protein level this means replaces arginine at residue 538 with cysteine — a missense variant. Submitter rationale: Variant summary: COL7A1 c.1612C>T (p.Arg538Cys) results in a non-conservative amino acid change located in the fibronectin type III domain (IPR003961) of the encoded protein sequence. Four of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 3.6e-05 in 251162 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.1612C>T in individuals affected with Dystrophic Epidermolysis Bullosa and no experimental evidence demonstrating its impact on protein function have been reported. One clinical diagnostic laboratory has submitted a clinical-significance assessment for this variant to ClinVar after 2014 without evidence for independent evaluation and classified the variant as likely benign. Based on the evidence outlined above, the variant was classified as uncertain significance.
