Uncertain significance for Autosomal recessive polycystic kidney disease — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_138694.4(PKHD1):c.12014A>C (p.Gln4005Pro), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glutamine with proline at codon 4005 of the PKHD1 protein (p.Gln4005Pro). The glutamine residue is weakly conserved and there is a moderate physicochemical difference between glutamine and proline. This variant is present in population databases (rs750272645, ExAC 0.001%). This variant has not been reported in the literature in individuals affected with PKHD1-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0"). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
