NM_001111.5(ADAR):c.3116A>G (p.Lys1039Arg) was classified as Uncertain significance for Symmetrical dyschromatosis of extremities; Aicardi-Goutieres syndrome 6 by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015). This variant lies in the ADAR gene (transcript NM_001111.5) at coding-DNA position 3116, where A is replaced by G; at the protein level this means replaces lysine at residue 1039 with arginine — a missense variant. Submitter rationale: This sequence change replaces lysine, which is basic and polar, with arginine, which is basic and polar, at codon 1039 of the ADAR protein (p.Lys1039Arg). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with clinical features of Aicardi-Gouti`eres syndrome and/or dyschromatosis symmetrica hereditaria (PMID: 16536805, 34631961). It has also been observed to segregate with disease in related individuals. ClinVar contains an entry for this variant (Variation ID: 2056173). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) has been performed for this missense variant. However, the output from this modeling did not meet the statistical confidence thresholds required to predict the impact of this variant on ADAR protein function. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
