Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000063.6(C2):c.1531G>A (p.Asp511Asn), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces aspartic acid, which is acidic and polar, with asparagine, which is neutral and polar, at codon 511 of the C2 protein (p.Asp511Asn). This variant is present in population databases (rs142802105, gnomAD 0.2%). This missense change has been observed in individual(s) with atypical hemolytic uremic syndrome (PMID: 32113979). Algorithms developed to predict the effect of missense changes on protein structure and function output the following: SIFT: "Tolerated"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0". The asparagine amino acid residue is found in multiple mammalian species, which suggests that this missense change does not adversely affect protein function.
