Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_005902.4(SMAD3):c.1231C>T (p.Leu411Phe), citing Ambry Variant Classification Scheme 2023: The p.L411F variant (also known as c.1231C>T), located in coding exon 9 of the SMAD3 gene, results from a C to T substitution at nucleotide position 1231. The leucine at codon 411 is replaced by phenylalanine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
