Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_001083116.3(PRF1):c.140G>A (p.Gly47Asp), citing LabCorp Variant Classification Summary - May 2015: Variant summary: PRF1 c.140G>A (p.Gly47Asp) results in a non-conservative amino acid change located in the Membrane attack complex component/perforin (MACPF) domain (IPR020864) of the encoded protein sequence. Algorithms developed to predict the effect of missense changes on protein structure and function all suggest that this variant is likely to be disruptive. The variant was absent in 246904 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.140G>A in individuals affected with Familial Hemophagocytic Lymphohistiocytosis and no experimental evidence demonstrating its impact on protein function have been reported. Other missense changes affecting the same residue (G47V/C) have been reported in individuals affected with hemophagocytic lymphohistiocytosis (HGMD), suggesting that this residue might be important for protein function. ClinVar contains an entry for this variant (Variation ID: 2054104). Based on the evidence outlined above, the variant was classified as uncertain significance.
