Uncertain significance for Hereditary spastic paraplegia 49 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_014844.5(TECPR2):c.1346C>T (p.Ser449Leu), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces serine, which is neutral and polar, with leucine, which is neutral and non-polar, at codon 449 of the TECPR2 protein (p.Ser449Leu). This variant is present in population databases (rs770524678, gnomAD 0.01%). This variant has not been reported in the literature in individuals affected with TECPR2-related conditions. ClinVar contains an entry for this variant (Variation ID: 2051822). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant¬†is likely to be tolerated.
