Pathogenic for Capillary malformation-arteriovenous malformation syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_002890.3(RASA1):c.539+4A>G, citing Invitae Variant Classification Sherloc (09022015): This sequence change falls in intron 1 of the RASA1 gene. It does not directly change the encoded amino acid sequence of the RASA1 protein. It affects a nucleotide within the consensus splice site. For these reasons, this variant has been classified as Pathogenic. Variants that disrupt the consensus splice site are a relatively common cause of aberrant splicing (PMID: 17576681, 9536098). Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. This variant has been observed in individual(s) with clinical features of RASA1-related conditions (Invitae). In at least one individual the variant was observed to be de novo. This variant is not present in population databases (gnomAD no frequency).
