This variant lies in the CHRNA2 gene (transcript NM_000742.4) at coding-DNA position 503, where C is replaced by T; at the protein level this means replaces threonine at residue 168 with methionine — a missense variant. Submitter rationale: Variant summary: CHRNA2 c.503C>T (p.Thr168Met) results in a non-conservative amino acid change located in the Neurotransmitter-gated ion-channel ligand-binding domain (IPR006202) of the encoded protein sequence. Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 2.4e-05 in 251444 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.503C>T in individuals affected with Autosomal Dominant Nocturnal Frontal Lobe Epilepsy 4 and no experimental evidence demonstrating its impact on protein function have been reported.
