Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Color Diagnostics, LLC DBA Color Health to NM_000059.4(BRCA2):c.517-14T>G, citing ACMG Guidelines, 2015. This variant lies in the BRCA2 gene (transcript NM_000059.4) at 14 bases into the intron immediately before coding-DNA position 517, where T is replaced by G. Submitter rationale: This variant causes a T to G nucleotide substitution at the -14 position of intron 6 of the BRCA2 gene. A splicing prediction algorithm suggests that this variant may weaken the intron 6 splice acceptor site (PMID: 30661751). To our knowledge, functional studies have not been reported for this variant. This variant has not been reported in individuals affected with hereditary cancer in the literature. This variant has been identified in 1/251096 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
