Uncertain significance for Tyrosinemia type I — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000137.4(FAH):c.1181G>A (p.Gly394Glu), citing Invitae Variant Classification Sherloc (09022015): In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive. This variant has not been reported in the literature in individuals affected with FAH-related conditions. This variant is present in population databases (rs370627856, gnomAD 0.007%). This sequence change replaces glycine, which is neutral and non-polar, with glutamic acid, which is acidic and polar, at codon 394 of the FAH protein (p.Gly394Glu).
