Uncertain significance for Charcot-Marie-Tooth disease axonal type 2U; Severe early-onset pulmonary alveolar proteinosis due to MARS deficiency — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_004990.4(MARS1):c.1705C>G (p.Leu569Val), citing Invitae Variant Classification Sherloc (09022015): In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive. This variant has not been reported in the literature in individuals affected with MARS-related conditions. This variant is not present in population databases (gnomAD no frequency). This sequence change replaces leucine, which is neutral and non-polar, with valine, which is neutral and non-polar, at codon 569 of the MARS protein (p.Leu569Val).
