Uncertain significance for Dilated cardiomyopathy 1R; Hypertrophic cardiomyopathy 11; Atrial septal defect 5 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_005159.5(ACTC1):c.61G>A (p.Ala21Thr), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the ACTC1 gene (transcript NM_005159.5) at coding-DNA position 61, where G is replaced by A; at the protein level this means replaces alanine at residue 21 with threonine — a missense variant. Submitter rationale: This sequence change replaces alanine, which is neutral and non-polar, with threonine, which is neutral and polar, at codon 21 of the ACTC1 protein (p.Ala21Thr). This variant is present in population databases (no rsID available, gnomAD 0.007%). This variant has not been reported in the literature in individuals affected with ACTC1-related conditions. ClinVar contains an entry for this variant (Variation ID: 2045899). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) has been performed for this missense variant. However, the output from this modeling did not meet the statistical confidence thresholds required to predict the impact of this variant on ACTC1 protein function. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
