NM_000393.5(COL5A2):c.3605G>A (p.Arg1202Gln) was classified as Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.R1202Q variant (also known as c.3605G>A), located in coding exon 50 of the COL5A2 gene, results from a G to A substitution at nucleotide position 3605. The arginine at codon 1202 is replaced by glutamine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
