NM_032043.3(BRIP1):c.2500A>G (p.Arg834Gly) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.R834G variant (also known as c.2500A>G), located in coding exon 17 of the BRIP1 gene, results from an A to G substitution at nucleotide position 2500. The arginine at codon 834 is replaced by glycine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
