Uncertain significance for Costello syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_005343.4(HRAS):c.177_433dup (p.Glu98Ter), citing Invitae Variant Classification Sherloc (09022015): This variant has not been reported in the literature in individuals affected with HRAS-related conditions. This variant is not present in population databases (gnomAD no frequency). This variant results in a tandem copy number gain of part of exon 3 and part of exon 4 of the HRAS gene. This variant is predicted to be out-of-frame, and may result in an absent or disrupted protein product. However, the current clinical and genetic evidence is not sufficient to establish whether loss-of-function variants in HRAS cause disease. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
