NM_000492.4(CFTR):c.3471G>A (p.Met1157Ile) was classified as Uncertain significance for Cystic fibrosis by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.M1157I variant (also known as c.3471G>A), located in coding exon 22 of the CFTR gene, results from a G to A substitution at nucleotide position 3471. The methionine at codon 1157 is replaced by isoleucine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
