Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_004944.4(DNASE1L3):c.41C>A (p.Ser14Tyr), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces serine, which is neutral and polar, with tyrosine, which is neutral and polar, at codon 14 of the DNASE1L3 protein (p.Ser14Tyr). This variant is present in population databases (rs142751723, gnomAD 0.007%). This variant has not been reported in the literature in individuals affected with DNASE1L3-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Tolerated"; PolyPhen-2: "Possibly Damaging"; Align-GVGD: "Class C0"). Experimental studies have shown that this missense change does not substantially affect DNASE1L3 function (PMID: 24206041).
