Uncertain significance for Townes syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_002968.3(SALL1):c.109A>T (p.Thr37Ser), citing Invitae Variant Classification Sherloc (09022015): This variant is present in population databases (no rsID available, gnomAD 0.007%). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. This variant has not been reported in the literature in individuals affected with SALL1-related conditions. This sequence change replaces threonine, which is neutral and polar, with serine, which is neutral and polar, at codon 37 of the SALL1 protein (p.Thr37Ser).
