This sequence change replaces arginine, which is basic and polar, with histidine, which is basic and polar, at codon 628 of the XYLT1 protein (p.Arg628His). This variant is present in population databases (rs775575069, gnomAD 0.005%). This variant has not been reported in the literature in individuals affected with XYLT1-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. Experimental studies have shown that this missense change does not substantially affect XYLT1 function (PMID: 16376579).
