Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000059.4(BRCA2):c.2264C>A (p.Ser755Tyr), citing Ambry Variant Classification Scheme 2023: The p.S755Y variant (also known as c.2264C>A), located in coding exon 10 of the BRCA2 gene, results from a C to A substitution at nucleotide position 2264. The serine at codon 755 is replaced by tyrosine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
