The variant was absent in 251482 control chromosomes. To our knowledge, no occurrence of c.661_713dup53 in individuals affected with STAR-related conditions and no experimental evidence demonstrating its impact on protein function have been reported. However, this variant disrupts a region of the STAR protein in which other variant(s) (e.g. c.824T>C, p.Leu275Pro; c.811del, p.Leu271fs) have been determined to be pathogenic internally. ClinVar contains an entry for this variant (Variation ID: 2039092). Based on the evidence outlined above, the variant was classified as pathogenic.
