Uncertain significance for Hereditary pancreatitis — the classification assigned by Ambry Genetics to NM_007272.3(CTRC):c.718C>T (p.Arg240Trp), citing Ambry Variant Classification Scheme 2023: The p.R240W variant (also known as c.718C>T), located in coding exon 7 of the CTRC gene, results from a C to T substitution at nucleotide position 718. The arginine at codon 240 is replaced by tryptophan, an amino acid with dissimilar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
