NM_001985.3(ETFB):c.160G>A (p.Glu54Lys) was classified as Likely pathogenic by GeneDx, citing GeneDx Variant Classification (06012015). This variant lies in the ETFB gene (transcript NM_001985.3) at coding-DNA position 160, where G is replaced by A; at the protein level this means replaces glutamic acid at residue 54 with lysine — a missense variant. Submitter rationale: p.Glu54Lys (GAG>AAG): c.160 G>A in exon 2 of the ETFB gene (NM_001985.2). An E54K variant that is likely pathogenic was identified in the ETFB gene. It has not been published as a mutation, nor has it been reported as a benign polymorphism to our knowledge. The E54K variant is a non-conservative amino acid substitution, which is likely to impact secondary protein structure as these residues differ in polarity, charge, size and/or other properties. This substitution occurs at a position that is highly conserved across species and in silico analysis predicts this variant is probably damaging to the protein structure/function. Therefore, this variant is a strong candidate for a pathogenic mutation, however the possibility that it is a benign variant cannot be excluded. The variant is found in MITONUC-MITOP panel(s).
