Uncertain significance for AVPR2-related condition — the classification assigned by PreventionGenetics, part of Exact Sciences to NM_000054.7(AVPR2):c.338G>T (p.Arg113Leu), citing ACMG Guidelines, 2015: The AVPR2 c.338G>T variant is predicted to result in the amino acid substitution p.Arg113Leu. To our knowledge, this variant has not been reported in the literature or in a large population database (http://gnomad.broadinstitute.org), indicating this variant is rare. Different substitutions affecting the same amino acid (p.Arg113Trp and p.Arg113Gln) have been reported to be causative for X-linked nephrogenic diabetes insipidus (Human Gene Mutation Database; http://www.hgmd.cf.ac.uk/). Although we suspect that this variant may be pathogenic, at this time, the clinical significance of this variant is uncertain due to the absence of conclusive functional and genetic evidence.
