Uncertain significance — the classification assigned by GeneDx to NM_000108.5(DLD):c.306T>A (p.His102Gln), citing GeneDx Variant Classification (06012015): p.His102Gln (CAT>CAA): c.306 T>A in exon 5 of the DLD gene (NM_000108.3) The H102Q missense substitution has not been published as a mutation, nor has it been reported as a benign polymorphism to our knowledge. The amino acid change is non-conservative in that Histidine is a positively charged amino acid and Glutamine is an uncharged amino acid. This change occurs at a position in the DLD protein that is not highly conserved. In-silico analyses are not consistent in their predictions of whether or not H102Q is damaging to the DLD protein. Therefore, based on the currently available information, it is unclear whether H102Q is a disease-causing mutation or a rare benign variant. The variant is found in MITONUC-MITOP panel(s).
