Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000430.4(PAFAH1B1):c.569-6T>C, citing Invitae Variant Classification Sherloc (09022015). This variant lies in the PAFAH1B1 gene (transcript NM_000430.4) at 6 bases into the intron immediately before coding-DNA position 569, where T is replaced by C. Submitter rationale: This variant has been observed in individual(s) with lissencephaly (Invitae). In at least one individual the variant was observed to be de novo. For these reasons, this variant has been classified as Pathogenic. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. This variant is not present in population databases (gnomAD no frequency). This sequence change falls in intron 6 of the PAFAH1B1 gene. It does not directly change the encoded amino acid sequence of the PAFAH1B1 protein.
