NM_002439.5(MSH3):c.654del (p.Lys219fs) was classified as Pathogenic by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. This variant has not been reported in the literature in individuals affected with MSH3-related conditions. This variant is present in population databases (rs779605105, gnomAD 0.0009%). This sequence change creates a premature translational stop signal (p.Lys219Asnfs*14) in the MSH3 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in MSH3 are known to be pathogenic (PMID: 27476653).
