Pathogenic for Spondylocostal dysostosis 3, autosomal recessive — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001040167.2(LFNG):c.216_217del (p.His72fs), citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. This variant has not been reported in the literature in individuals affected with LFNG-related conditions. This variant is not present in population databases (gnomAD no frequency). This sequence change creates a premature translational stop signal (p.His72Glnfs*78) in the LFNG gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in LFNG are known to be pathogenic (PMID: 16385447, 29459493).
