This variant lies in the ALMS1 gene (transcript NM_001378454.1) at coding-DNA position 786 through coding-DNA position 788, duplicating 3 bases. Submitter rationale: The c.789_791dupAGA variant (also known as p.E263dup), located in coding exon 5 of the ALMS1 gene, results from an in-frame duplication of AGA at nucleotide positions 789 to 791. This results in the duplication of an extra residue between codons 263 and 264. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive (Choi Y et al. PLoS ONE. 2012; 7(10):e46688). Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
