Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_007194.4(CHEK2):c.744A>G (p.Ile248Met), citing Ambry Variant Classification Scheme 2023: The p.I248M variant (also known as c.744A>G), located in coding exon 5 of the CHEK2 gene, results from an A to G substitution at nucleotide position 744. The isoleucine at codon 248 is replaced by methionine, an amino acid with highly similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
